This in-depth course covers the design, operational principles and practical application of modern instrumental methods used in chemical analysis. These methods are used in the separation, identification and quantification of the chemical components of natural and artificial materials. Using a combination of problem-based learning approaches, case studies and traditional lectures, the student will develop critical thinking skills in the areas of instrument selection, method development and data interpretation.

The lecture program will address issues related to the discussion of sampling and storage procedures used for chemical analysis. The steps of methods development and their statistical assessment, like validation of the measurement procedure in terms of quality (calibration, sensitivity, precision and accuracy of measurement, selectivity and specificity of methods). In this case, the aspects of sample pretreatment methods used during sample clean-up of complex sample matrices (including solid phase extraction procedures, liquid membrane extraction, solvent extraction, etc.) will be also presented. The theoretical concept and analytical application of chromatographic techniques, including gas chromatography, liquid chromatography and supercritical fluid chromatography, as well as electromigration techniques (especially capillary electrophoresis) will be discussed too. Furthermore, the issues related to the use of atomic and emission spectroscopy, infrared (IR), X-ray and nuclear resonance spectrometry (NMR), mass spectrometry, in the area of chemical analysis , and the topics of scanning electron microscopy, thermal analysis, and the use of combined techniques (like LC-NMR-MS, GC-IR-MS, GCxGC, LCxLC, etc.)will be also presented.

B. Laboratory issues:

Laboratory classes are aimed at acquiring practical knowledge in working with analytical equipment commonly used in analytical laboratories such as HPLC, and CE using UV-VIS and MS detection, as well as the use of IR and NMR spectroscopy in solid and liquid samples. The techniques of extraction procedures (SPE, LLE, UAE, etc) used for sample clean-up step. Acquiring the ability to choose the method and type of apparatus to perform a specific analytical determination.

Example of performed experiments:

1. Separation and determination of selected antidepressants using capillary electrophoresis system with UV detection.

2. Isolation, preconcentration and determination of dyes from spice sample (chilli) using solid-phase extraction and HPLC-UV-VIS technique.

3. Enancioseparation of amino acids using capillary electrophoresis.

4. Isolation of theobromine from cocoa and product identification via HPLC analysis

5. FT-IR analysis for identification of plastics and organic compounds.

6. NMR technique for structure identification of organic compounds.

7. Isolation of nicotine from tobacco and urine samples and its analysis with LC-MS technique

Students will gain the necessary knowledge of the principles ofinstrumental scientific research. Theknowledgewill include the design and analysis techniques of mass spectrometry, ion mobility, high performance liquid chromatography, ultraviolet-visible spectrophotometryand fluorescence. 1: Graduate students should demonstrate expertise (breadth and depth) in providing a thorough understanding of the chemical principles related to this range of instrumental techniques.2:Graduate students should demonstrate an ability to work independentlyand complete assignments.3: Graduate students should demonstrate the ability to assimilate and disseminate the scientific knowledgethrough assignments and exams.

Metody i kryteria oceniania: (tylko po angielsku)

There will be 10 labs which cover each section of this course. In the laboratory, students will gain hands­on experience of instrumental analysis by performing a series of chemical determinations in compelx sample matrices. Students will prepare his/her own written report which is written in a professional format including title, abstract, introduction, experimental section, results, discussion, conclusions, acknowledgement and references.

This course builds on students' existing background in analytical chemistry to develop both theory and practice relating to the latest analytical techniques used in industry and research.
